摘  要:文中以贺兰山上的耐旱乔木灰榆的根际土壤为研究材料,在对其根际细菌进行分离纯化的基础上采用PEG6000模拟干旱环境进行筛选,得到一株耐旱菌TH11。进一步采用10%~80%的PEG6000模拟不同干旱程度培养菌株TH11以探究其耐旱能力,在明确其生长特性的基础上扩增其16S r DNA序列并构建系统发育树以确定该菌株的分类地位。结果表明:菌株TH11归属于虚构芽孢杆菌(Fictibacillus),其在p H值7. 5、37℃条件下生长最好;此培养条件下测得该菌在0~4h为调整期,4~45h为对数生长期,45~80h为生长稳定期,80h以后为衰亡期;其生物量大且能够耐受80%PEG6000的干旱胁迫,具有较强的耐旱能力。In order to obtain drought tolerant bacteria,the rhizosphere soil of Ulmus glaucescens in Helanshan Mountain was collected for strain isolation. Based on isolation and purification of rhizosphere bacteria,screening of drought tolerant bacteria was developed by using PEG simulating drought circumstance,and strain TH11 was screened as the most drought tolerant bacterium. To make clear the drought tolerance of strain TH11,further researched was designed by adding different concentrations of PEG6000 into the medium to get different drought stress levels. After the growth characteristics of strain TH11 was studied,the 16 S r DNA was sequenced and the phylogenetic tree of 16 S r DNA was constructed to determine the taxonomic status of strain TH11. Results showed that strain TH11 belongs to Fictibacillus sp.,with the optimum growth pH value and temperature being 7. 5 and37℃,respectively. Under the optimal conditions of strain TH11,the adjustment period,the logarithmic growth period,and the growth stability period were 0 ~ 4 h,5 ~ 45 h,45 ~ 80 h,respectively. According to the growth curve,80 h was the critical point between the growth stability period and the decline period. Strain TH11 was able to tolerate 80% PEG600 stress and showed strong drought tolerance.
