微乳薄层色谱法对桑叶桑枝中活性成分的鉴别

摘  要:目的建立微乳薄层色谱法鉴别桑叶桑枝中活性成分的方法。方法85%乙醇为溶剂,超声提取桑叶、桑枝中的活性成分。以聚酰胺薄膜作为固定相,微乳液为流动相,对桑叶桑枝中的活性成分进行定性鉴别,并考察微乳液的含水量、十二烷基硫酸钠(SDS)用量、助表面活性剂的种类及用量、微乳液中甲酸含量对分离效果的影响。结果最佳展开系统为含水量75%的O/W型微乳液,成分和配比为m(SDS)∶m(正丁醇)∶m(正己烷)2.7∶6.3∶1.0,V(微乳液)∶V(甲酸)9.0∶1.0。分别鉴定出桑叶中含有绿原酸、芦丁和桑皮苷A3种活性成分,桑枝中含有桑辛素、东莨菪内酯、绿原酸、桑皮苷A4种活性成分。结论本实验快速简单、点样量小、斑点清晰、重复性好,适用于桑叶桑枝中活性成分的鉴别。Objective To establish a microemulsion thin layer chromatography method for identifying active components in Folium mori and Ramulus mori.Methods The active constituents of Folium mori and Ramulus mori were extracted by ultrasonic method using 85%ethanol as solvent.The active components were identified qualitatively using the polyamide film as stationary phase and the microemulsion as mobile phase.The effect of water content in microemulsion,amount of SDS and n-butanol,type and dosage of cosurfactant,and formic acid content of microemulsion on separation were investigated.Results The optimal development system was an O/W microemulsion with 75%water content,The composition and ratio were m(sodium dodecyl sulfate)∶m(n-butanol)∶m(n-hexane)2.7∶6.3∶1.0,V(microemulsion)∶V(formic acid)9.0∶1.0 respectively.Three components of chlorogenic acid,rutin and mulberroside A in Folium mori and four components of morusin,scopoletin,chlorogenic acid,and mulberroside A in Ramulus mori were identified.Conclusion This experiment is fast,simple,small sample size,clear spots,and good repeatability,which is suitable for identification of active ingredients in Folium mori and Ramulus mori.
