冬凌草甲素对棕榈酸诱导的巨噬细胞炎症反应的影响

摘  要:目的研究冬凌草甲素对棕榈酸刺激的RAW264.7小鼠巨噬细胞炎症反应的影响。方法体外培养RAW264.7细胞,噻唑蓝比色法(Mr/T)确定冬凌草甲素作用的最佳浓度及时间;将细胞分为空白对照组、棕榈酸组(500μmol/L棕榈酸,刺激细胞24h)、冬凌草甲素预处理组(冬凌草甲素预处理细胞24h后,再给予500wmol/L棕榈酸刺激细胞24h),酶联免疫吸附试验(ELISA)法检测上清液中肿瘤坏死因子(TNF)-d、白细胞介素(IL)-6、IL-10的浓度,观察细胞的炎症状态;实时定量聚合酶链反应(RT-PCR)分析RAW264.7细胞TNF-α、IL-6、IL-10mRNA的表达水平。结果冬凌草甲素作用于RAW264.7细胞的最佳浓度为5μmol/L,时间为24h。棕榈酸组RAW264.7细胞内炎症因子TNF-α、IL-6mRNA表达水平及细胞上清中TNF-α、IL-6的浓度明显高于空白对照组(P均〈0.05),IL-10则明显低于空白对照组(P〈0.05)。与棕榈酸组比较,冬凌草甲素预处理组RAW264.7细胞内炎症因子TNF-α、IL-6mRNA表达水平及细胞上清中TNF-α、IL-6的浓度明显低于棕榈酸组(P均〈0.05),IL-10则明显高于棕榈酸组(P〈0.05)。结论冬凌草甲素能抑制棕榈酸诱导的RAW264.7细胞炎症因子的表达,减轻其炎症反应。Objective To investigate the effect of oridonin in palmitic acid-induced inflammatory response of macrophages(RAW264.7cells). Methods The optimum time and concentration of oridonin on RAW264.7 were determined by MTT assay. The models of RAW264.7 were established and divided into 3 groups, control group, palmitic acid group and oridonin group. In the oridonin group, RAW264.7 cells were pretreated with oridonin under the condition of 500 μmol/L palmitic acid, while other groups palmitic acid group received 500 μmol/L paimitic acid. After 24 h treatment, the concentration of tumor necrosis factor-or (TNF-ct), interleukin-6 (IL-6) and IL-10 in the supernatant were detected by the enzyme-linked immunosorbent assay(ELISA) to observe the inflammation state, while the mRNA expression of TNF-α,IL-6 and IL-10 were detected by the real time polymerase chain reaction (PCR). Results The optimum concentration of oridonin was 5 μmol/L, and the optimum time was 24 h. Compared with the control group,the mRNA expression of TNF-α and IL-6 in RAW264.7 cells and the concentration of TNF-α and IL-6 in the supernatant were significantly higher, while IL-10 was significantly lower ( all P 〈 0.05 ) in palmitic acid group. But in the oridonin group, oridonin significantly induced the expression of IL-10 mRNA in RAW264.7 cells and the concentration of it in the supernatant ( all P 〈 0.05 ). But the mRNA expression of TNF-α, IL-6 in RAW264. 7 cells and the concentration of them in the supernatant significantly reduced ( all P 〈 0. 05 ). Conclusion Oridonin can inhibit the expression of cytokines induced by palmitic acid in RAW264.7 cells and alleviate the inflammatory state of the cells.
