摘  要:目的对近年来新出现的三-(2,3-二溴丙基)异氰脲酸酯(TBC)的慢性毒性进行研究。方法试验采用清洁级的Wistar大鼠80只,雌雄各半,随机分成四组,1.250g/(kg·bw)、0.625g/(kg·bw)、0.313g/(kg·bw)三个染毒剂量和对照组。每个剂量组20只大鼠,连续经口染毒28天后停止染毒,各脏器称重,计算脏器体重比,腹腔取血进行生化和血液检查,同时对心、肝、脾、肺、肾进行病理学检查。结果染毒组和对照组的肝/体比差异有统计学意义且有剂量效应关系(雄高t=2.035,P<0.05;雌高t=3.169,P <0.01);雌性染毒组与对照组的肾/体比差异有统计学意义(雌高t=4. 300,P <0.01;雌中t=5. 225,P <0.01)。高剂量组心肌纤维紊乱排列,心肌细胞扭曲、肥大,细胞间隙较大,同时伴有炎症浸润。肝的病理片中,随着染毒剂量的增加,肝细胞逐渐发生肿胀,颗粒变性,肝索结构不清楚,间质成分增生,肝血窦增宽,肺泡出现不均匀扩张,肺泡壁明显增厚,毛细血管内皮细胞肿胀明显,可见中性粒子细胞聚集,亦可见充血和水肿以及炎性渗出。肾组织中水肿明显,且炎细胞浸润,间质有散落出血点,且结构模糊。结论 TBC毒性作用的靶器官可能是肝、肺、肾,且可能引起贫血和高血脂,而雌鼠比雄鼠对TBC的中毒反应明显。Objective To study the chronic toxicity of tris( 2,3-dibromopropyl) isocyanurate( TBC) in recent years. Methods Eighty Wistar rats were randomly divided into four groups: 1. 250 g/( kg·bw),0. 625 g/( kg·bw),0. 313 g/( kg·bw) three dose groups and control group. After 28 days exposure,the organ weight ratio were calculated,peritoneal blood was taken for biochemical and hematological examinations,also the pathological examination of the heart,liver,spleen,lung and kidney. Results There were significant differences between exposure group and the control group of liver/body and dose effect relationship exist( male high t = 2. 035,P < 0. 05;female high t = 3. 169,P < 0. 01);there were significant differences of kidney/body female dose group compared with the control group( female high t = 4. 300,P < 0. 01;female medium t = 5.225,P < 0. 01). Pathological examination of heart,liver,spleen,lung and kidney showed that myocardial fibers were arranged in disorder in high dose group,cardiac muscle cells were distorted,hypertrophic,and had large cell gap,accompanied by inflammatory infiltration. The pathological sections of liver,as the dose increased,liver cells gradually swelling,granular degeneration,hepatic cord structure were not clear,stroma hyperplasia,liver sinusoids widened,uneven alveolar expansion,alveolar wall thickening,capillary endothelial cell swelling significantly,neutral cell aggregation,also visible hyperemia,as well as inflammatory exudation. The edema was obvious in the renal tissue,and the inflammatory cells were infiltrated. The interstitium was scattered and bleed,and the structure was blurred. Conclusion The target organs of TBC may be liver,lung,kidney and TBC may cause anemia and high blood lipids,while the male rats were more sensitive on TBC poisoning.
